Biological Background: NPY Function and Localization
- Pro-neuropeptide Y (NPY) is a member of the neuropeptide Y family, produced from a precursor protein through post-translational cleavage into active peptides, including the mature NPY hormone.
- NPY acts as a ligand for neuropeptide Y receptor type 1 (NPY1R) and type 2 (NPY2R). Receptor binding triggers adenylate cyclase-inhibiting G protein-coupled receptor signaling, resulting in decreased cAMP production, and also increases intracellular Ca(2+) levels. - NPY is involved in the regulation of eating behavior, acting as a potent stimulator of food intake.
- In the gastrointestinal system, NPY contributes to the regulation of colonic antisecretory tone and contractility.
- Subcellularly, NPY is secreted and stored in cytoplasmic vesicles, particularly in neuronal dense core vesicles.
- One of the most abundant peptides in the nervous system, NPY is also found in some chromaffin cells of the adrenal medulla.
