Biological Background: NPRL2 Function and Localization

  • NPRL2 (also known as NPR2, NPR2L, TUSC4, FFEVF2) is a catalytic component of the GATOR1 complex, which acts as an inhibitor of the amino acid-sensing branch of the mTORC1 pathway. Related references: PMID:23723238, PMID:29590090, PMID:35338845
  • Upon amino acid deprivation, GATOR1 acts as a GTPase-activating protein (GAP) for RagA/RRAGA or RagB/RRAGB, converting them to the inactive GDP-bound state, leading to mTORC1 release from lysosomes and pathway inhibition. Related references: PMID:23723238, PMID:29590090, PMID:35338845
  • When amino acids are abundant, GATOR2 ubiquitinates and inhibits GATOR1, allowing mTORC1 activation; additionally, PRMT1 methylates NPRL2 under methionine-sufficient conditions, inhibiting its GAP activity.   • Validated applications include Western blot (WB), immunofluorescence/immunocytochemistry (IF/ICC), and ELISA. Cross-reactivity with human, mouse, and rat samples has been reported; consider species-specific validation for your experimental system.
  • For WB, a band at approximately 44 kDa is expected. Due to the polyclonal nature, additional non-specific bands may be observed; optimization of blocking and antibody dilution is recommended.
  • When using IF/ICC, appropriate fixation and permeabilization methods should be tested, as NPRL2 localizes to lysosomal membranes.
