Biological Background: NPR3 Function and Localization
- Encodes the atrial natriuretic peptide receptor 3 (NPR3), also known as the clearance receptor (NPR-C / ANP-C), which binds natriuretic peptides NPPA (ANP), NPPB (BNP), and NPPC (CNP) with comparable affinity.
- Functions primarily as a clearance receptor, internalizing and degrading circulating natriuretic peptides to regulate their local concentrations and downstream signaling effects.
- Acts as a modulator of bone growth and osteoblast differentiation by binding the ligand osteocrin; this interaction prevents natriuretic peptide binding and leads to increased cGMP production.
- Localizes to the cell membrane as a single-pass transmembrane protein, with a large extracellular ligand-binding domain and a short cytoplasmic tail.
- Contains structural features typical of the natriuretic peptide receptor family, including a signal peptide, multiple disulfide bonds, and N-linked glycosylation sites.
- Alternative splicing generates multiple transcript variants, and mutations in this gene have been associated with disease phenotypes.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 272–541 of human NPR3 (isoform NP_001191304.1), which covers a portion of the extracellular domain; consider this when designing detection/blocking strategies.
- For Western blot, a band near 60 kDa is expected; we recommend titrating the antibody (e.g., 1:500–1:2000) and validating it in relevant human, mouse, or rat tissue lysates.
- In ELISA applications, the polyclonal nature may provide enhanced sensitivity; consider pairing with a monoclonal capture antibody if a sandwich format is desired.
