Biological Background: NPLOC4 Function and Localization
- NPLOC4 (Nuclear Protein Localization Protein 4 Homolog), also known as NPL4, is a component of the UFD1-VCP-NPLOC4 ternary complex that recognizes ubiquitinated proteins and mediates their export from the endoplasmic reticulum to the cytoplasm for proteasomal degradation.
- The NPLOC4-UFD1-VCP complex regulates spindle disassembly at the end of mitosis and is required for closed nuclear envelope formation (By similarity).
- Acts as a negative regulator of type I interferon production by forming a complex with VCP and UFD1 that binds RIGI, recruiting RNF125 to promote ubiquitination and degradation of RIGI. Related references: PMID:26471729
- Localizes to the cytoplasm, cytosol, endoplasmic reticulum, and nucleus, supporting roles in both ER-associated degradation and nuclear processes.
- Highest expression is observed in brain, heart, skeletal muscle, kidney, and fetal liver.
- Contains a zinc-finger domain and undergoes acetylation; also subject to alternative splicing. Functional keywords include chaperone, Ubl conjugation pathway, and reference proteome.

- In immunocytochemistry/immunofluorescence (IF/ICC), optimize fixation and permeabilization to detect NPLOC4 in its various subcellular locations (ER, cytosol, nucleus).
