Biological Background: NPL Function and Localization
- N-acetylneuraminate lyase (NPL), also known as sialic acid aldolase or sialate lyase, is a cytoplasmic enzyme encoded by the NPL gene. It belongs to the lyase family and catalyzes the cleavage of N-acetylneuraminic acid (sialic acid) to pyruvate and N-acetylmannosamine via a Schiff base intermediate. Related references: PMID:33895133
- By breaking down sialic acids, NPL prevents their recycling back to the cell surface, playing a crucial role in regulating sialic acid levels on glycoproteins and glycolipids. Related references: PMID:33895133
- NPL is involved in the degradation pathway of N-glycolylneuraminic acid (Neu5Gc), a non-human sialic acid obtained from the diet that must be cleared from the body. Related references: PMID:22692205, PMID:33895133
- The protein localizes to the cytoplasm and is expressed in multiple tissues; isoform 2 is specifically found in placenta, liver, kidney, pancreas, spleen, thymus, ovary, small intestine, and peripheral blood leukocytes.
- The mature protein has a molecular weight of approximately 35 kDa, consistent with the product specification of 35kDa.
- Alternative splicing generates multiple transcript variants, and the enzyme functions via a Schiff base intermediate, classified under carbohydrate metabolism and lyase activity.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering amino acids 1-250 of human NPL; this N-terminal region may be useful for detecting full-length NPL in WB. Consider reduced denaturing conditions given the expected band near 35 kDa.
- Cross-reactivity with mouse and rat suggests the antibody may be suitable for studies in rodent models; validate expression in relevant tissues such as liver or kidney where NPL is known to be expressed.
- For ELISA, the polyclonal nature may provide robust signal capture; however, titration and validation in the relevant sample matrix (e.g., cell lysates, tissue homogenates) are essential.
