Biological Background: NPC2 Function and Localization
- NPC2, also known as epididymal secretory protein E1 (HE1), is a small glycoprotein critical for intracellular cholesterol trafficking and lysosomal cholesterol export.
- NPC2 acts as an intracellular cholesterol transporter, facilitating cholesterol egress from lysosomes in concert with NPC1. Related references: PMID:11125141 PMID:15937921 PMID:17018531
- Unesterified cholesterol derived from LDLs is transferred by NPC2 to the N-terminal cholesterol-binding pocket of NPC1 for subsequent export. Related references: PMID:17018531 PMID:18772377 PMID:27238017
- NPC2 binds cholesterol with a 1:1 stoichiometry and can also interact with various sterols, including plant sterols such as stigmasterol and β-sitosterol. Related references: PMID:17018531
- A secreted form of NPC2 regulates biliary cholesterol secretion through stimulation of ABCG5/ABCG8-mediated cholesterol transport.

- The antibody is raised against full-length recombinant human NPC2 protein; optimal performance is expected in western blot detecting the ~17 kDa band.
- For western blot, consider using a 12% or 15% SDS-PAGE gel to resolve the 17 kDa NPC2 protein.
- Immunofluorescence and immunohistochemistry of endogenous NPC2 may require permeabilization and antigen retrieval steps; validate the protocol in your model system.
