Biological Background: NPC2 Function and Localization
- NPC2 (also known as HE1, EDDM1) is an intracellular cholesterol transporter that plays a critical role in the egress of cholesterol from the lysosomal compartment.
- It acts in concert with NPC1; cholesterol released from LDLs in late endosomes/lysosomes is transferred by NPC2 to the cholesterol-binding pocket of NPC1.

- The protein contains a signal peptide and features post-translational modifications such as disulfide bonds, glycosylation, and acetylation; mutations in NPC2 are linked to Niemann-Pick disease type C2.

- The antibody was raised against a recombinant fusion protein spanning amino acids 20–151 of human NPC2, which corresponds to the full mature protein after signal peptide cleavage. This design may ensure recognition of endogenous NPC2.
- For Western blotting, anticipate a band at approximately 17 kDa; validate the antibody in positive control lysates (e.g., human fibroblasts or kidney tissue) and consider using a reducing agent given the presence of disulfide bonds.
- In IHC-P or IF-P applications, include appropriate antigen retrieval steps and verify staining on tissue sections known to express NPC2 (such as kidney or testis).
- The antibody cross-reacts with mouse and rat NPC2; however, sequence homology between species should be checked for specific epitopes, and the antibody should be validated in the relevant experimental model.
