Biological Background: NOXA2/p67phox (NCF2) Function and Localization
- Neutrophil cytosol factor 2 (NCF2), also known as NOXA2, p67-phox, p67-PHOX, and 67 kDa neutrophil oxidase factor, is a cytosolic subunit of the phagocyte NADPH oxidase complex.
- NCF2 is essential for superoxide anion (O2(-)) production by mediating electron transfer from NADPH to O2 within the activated NADPH oxidase complex (PubMed:12207919, PubMed:38355798). Related references: PMID:12207919, PMID:38355798
- Activation of the NADPH oxidase complex is initiated by phosphorylation-dependent binding of the cytosolic NCF1/p47-phox subunit to CYBA/p22-phox, promoting assembly at the plasma membrane or phagosomal membrane.
- The protein localizes to the cytoplasm and translocates to membranes upon activation.
- NCF2 is a phosphoprotein and contains TPR repeats and an SH3 domain, which mediate protein-protein interactions within the oxidase complex.
- Mutations in NCF2 are associated with chronic granulomatous disease, a primary immunodeficiency characterized by defective superoxide production.
- Alternative splicing generates multiple isoforms, and the canonical isoform has a molecular weight of approximately 60 kDa.
