Biological Background: NOX1 Function and Localization
- NOX1, also known as MOX1 (mitogenic oxidase 1), NOH-1, or GP91-2, is a member of the NADPH oxidase family of enzymes that generate reactive oxygen species (ROS).
- The primary function of NOX1 is to transfer electrons from NADPH to molecular oxygen, generating superoxide anions. This activity requires cofactors including FAD, heme, and iron.
- The NOX1 gene is predominantly expressed in the colon, uterus, and prostate, and has been detected in colon carcinoma cells, implicating its role in cell proliferation and cancer biology.
- NOX1 localizes to the cell membrane and invadopodium membrane, a specialized cell projection involved in extracellular matrix degradation, linking its ROS production to cell invasion and migration.
- As indicated by its alternative name "mitogenic oxidase 1", NOX1-mediated superoxide production can promote mitogenic signaling pathways, potentially contributing to tumorigenesis in colon and prostate cancers.
- Post-translational modifications of NOX1 include phosphorylation and glycosylation, which may regulate its activity and localization.
- The human NOX1 gene produces multiple alternatively spliced transcript variants, which may contribute to the functional diversity and regulation of NOX1 activity in different tissues.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the central region of human NOX1 (aa 200-300). Consider this when assessing isoform specificity or if using the antibody for detection of alternatively spliced variants.
- For Western blotting, the predicted molecular weight is approximately 65 kDa; however, glycosylation may cause an upward shift. Validate the antibody in the target tissue or cell line.
- In immunofluorescence (IF/ICC), expect membrane and invadopodium staining. Optimize fixation and permeabilization to preserve membrane structures.
- For ELISA, use the immunogen peptide as a positive control to evaluate antibody titer and specificity.
- Cross-reactivity has been confirmed only for human. Validate in other species if needed before use.
