Biological Background: NOTCH4 Function and Localization

  • NOTCH4 (Neurogenic locus notch homolog protein 4) is a member of the Notch family of transmembrane receptors, which are key regulators of cell-fate determination.
  • It functions as a receptor for membrane-bound ligands Jagged1, Jagged2 and Delta1, mediating short-range cell-cell communication.
  • Upon ligand binding, the intracellular domain (NICD) is cleaved and translocates to the nucleus, where it forms a transcriptional activator complex with RBPJ/RBPSUH to activate target genes.
  • NOTCH4 signaling modulates diverse cellular processes including differentiation, proliferation, and apoptosis, with a potential role in vascular branching morphogenesis (by similarity).
  • The protein is localized at the cell membrane and, upon activation, in the nucleus.
  • NOTCH4 expression is tissue-specific: highly expressed in heart, moderately in lung and placenta, and at low levels in several other tissues; it is absent from adult brain and peripheral blood leukocytes.
  • Structurally, NOTCH4 contains multiple EGF-like repeats, ANK repeats, and other domains typical of Notch receptors; it undergoes post-translational modifications including glycosylation and phosphorylation.

Experimental Guidance and Technical Tips

  • The immunogen used is a recombinant protein spanning the C-terminal region (aa 1824-2003) of human NOTCH4, which may detect both full-length and cleaved intracellular domain.
  • For Western blotting, a band of ~210 kDa corresponding to full-length NOTCH4 may be observed; however, due to proteolytic processing, additional lower molecular weight bands might appear. Validation in the relevant sample system is recommended.
  • In immunocytochemistry/immunofluorescence (IF/ICC), anticipate membrane localization in unstimulated cells and both membrane and nuclear staining following ligand activation.
  • The antibody is also suitable for ELISA applications for detection or capture of NOTCH4.
  • Cross-reactivity with mouse and rat has been reported, but performance should be verified under specific experimental conditions.
