Biological Background: NOTCH2 Function and Localization
- NOTCH2 (Neurogenic locus notch homolog protein 2), also known as AGS2, HJCYS, hN2, is a large single-pass type I transmembrane receptor that plays a central role in the Notch signaling pathway, governing cell-fate decisions.
- Upon engagement by membrane-bound ligands Jagged-1, Jagged-2, or Delta-1, the intracellular domain (NICD) is released and translocates to the nucleus, where it forms a transcriptional activator complex with RBPJ to induce target genes such as the enhancer of split locus. - NOTCH2 belongs to the NOTCH receptor family, characterized by multiple extracellular EGF-like repeats and intracellular ankyrin (ANK) repeats, both essential for ligand binding and signal transduction.
- Subcellularly, NOTCH2 localizes at the cell membrane; following activation, NICD enters the nucleus, and the protein is also detected in the cytoplasm.
- The protein is subject to glycosylation and phosphorylation, containing disulfide bonds and being a phosphoprotein; it is classified under transcription regulation, developmental protein, and glycoprotein according to UniProt keywords.
- NOTCH2 is widely expressed, with high levels in brain, heart, kidney, lung, skeletal muscle, and liver, and shows ubiquitous expression during embryonic development.

Experimental Guidance and Technical Tips
- The immunogen spans amino acids 1697–1819 within the cytoplasmic region of human NOTCH2. This sequence is highly conserved across human, mouse, and rat, supporting broad reactivity.
- For Western blotting, expect a band near 265 kDa for full-length NOTCH2; cleavage products may appear at lower molecular weights. Validation in NOTCH2-expressing cell lines (e.g., liver cancer or osteoclast models) is advisable.
- In immunofluorescence/ICC, epitope accessibility may require permeabilization for intracellular detection; nuclear localization can be visualized under appropriate ligand-activation conditions.
- When used in ELISA, the antibody can serve as a capture or detection reagent for human, mouse, or rat NOTCH2, but optimal coating concentrations should be determined empirically.
