Biological Background: NOTCH2 Function and Localization
- Neurogenic locus notch homolog protein 2 (NOTCH2) is a receptor in the Notch signaling pathway, critical for cell-fate determination, differentiation, proliferation, and apoptotic programs (By similarity).
- NOTCH2 is activated by membrane-bound ligands Jagged-1 (JAG1), Jagged-2 (JAG2), and Delta-1 (DLL1), leading to release of the Notch intracellular domain (NICD) that forms a transcriptional activator complex with RBPJ/RBPSUH to regulate target genes (PubMed:21378985, PubMed:21378989). - For immunofluorescence/immunocytochemistry (IF/ICC), optimize fixation and permeabilization conditions, as NOTCH2 can localize to multiple subcellular compartments.
- In ELISA, titrate the antibody in a direct or capture format and evaluate performance against a standard curve of the immunogen or a relevant NOTCH2 fragment.
