Biological Background: Notch1 Function and Localization

  • Notch1 functions as a receptor for membrane-bound ligands Jagged-1, Jagged-2, and Delta-1, regulating cell-fate determination through the canonical Notch signaling pathway.
  • Upon ligand activation, the Notch intracellular domain (NICD) is released and translocates to the nucleus to form a transcriptional activator complex with RBPJ/RBPSUH, driving expression of enhancer of split locus genes.
  • Affects the implementation of differentiation, proliferation, and apoptotic programs; negatively regulates endothelial cell proliferation and migration, thereby contributing to angiogenesis control.
  • Important for the maturation of both CD4+ and CD8+ cells in the thymus and for follicular differentiation, possibly influencing cell fate selection within the follicle.
  • During cerebellar development, acts as a receptor for neuronal DNER and is involved in the differentiation of Bergmann glia.
  • Represses neuronal and myogenic differentiation; may play an essential role in postimplantation development, mesoderm development, somite formation, and neurogenesis.
  • Localizes to the cell membrane, late endosome membrane, and nucleus (after cleavage and nuclear translocation of NICD).
  • In fetal tissues, most abundant in spleen, brain stem, and lung; in adult tissues, found mainly in lymphoid tissues.

Experimental Guidance and Technical Tips

  • When using this antibody for western blot, consider that the full-length Notch1 has a predicted molecular weight of approximately 273 kDa, but cleavage products may also be observed. Validate the band pattern in your specific sample system.
  • For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval and fixation conditions may influence staining. Optimize protocols using appropriate positive and negative controls.
  • The immunogen corresponds to amino acids 2428–2555 in the C-terminal region of human Notch1; the antibody may recognize both full-length and cleaved forms depending on sample preparation.
  • Cross-reactivity with mouse and rat Notch1 is indicated; however, we recommend verifying reactivity and specificity in the relevant species and tissue context.
