Biological Background: NOS1 Function and Localization
- Encodes nitric oxide synthase 1, the neuronal isoform that produces nitric oxide (NO), a versatile signaling molecule acting as a neurotransmitter in the central and peripheral nervous systems.
- Belongs to the NOS family of enzymes, which also includes NOS2 (inducible) and NOS3 (endothelial), and is characterized as constitutive and calcium/calmodulin-dependent.
- Localizes to the sarcolemma, cell membrane, and dendritic spines, positioning it at synapses for rapid NO-mediated signaling.
- Mediates protein S-nitrosylation of cysteine residues on cytoplasmic targets, such as SRR, thereby modulating protein function and redox-sensitive pathways.
- Ubiquitously expressed: isoform 1 is found in brain, skeletal muscle, testis, lung, kidney, and at low levels in heart, adrenal gland, and retina; isoform 3 is testis-specific; isoform 4 is present in testis, skeletal muscle, lung, kidney.
- Contains multiple cofactor-binding domains (FAD, FMN, NADP, heme, iron) and a calmodulin-binding site, and is regulated by phosphorylation and ubiquitination.

- The immunogen corresponds to the N-terminal region (amino acids 1-180), which may recognize multiple isoforms due to alternative splicing; consider verifying isoform specificity in your experimental system.
- For Western blot, whole cell lysates from human, mouse, or rat brain or skeletal muscle tissues are appropriate positive sample choices.
- In immunofluorescence/immunocytochemistry, expect staining at the plasma membrane and dendritic spines; permeabilization may be required for intracellular epitopes.
- As a polyclonal antibody, it may exhibit batch-to-batch variation; include appropriate controls and validate performance in your specific application.
