Biological Background: Catenin Beta-1 (β-Catenin) Function and Localization

Serves as a key downstream component of the canonical Wnt signaling pathway. In the absence of Wnt, β-catenin is phosphorylated by a destruction complex (AXIN1, APC, GSK3B, CSNK1A1) and targeted for proteasomal degradation. Upon Wnt stimulation, β-catenin is stabilized, accumulates in the nucleus, and functions as a coactivator for TCF/LEF transcription factors to activate Wnt-responsive genes.

Also acts as a transcriptional coactivator for other factors such as NR5A2, expanding its regulatory roles beyond canonical Wnt signaling.

- Since this antibody is specific for the non-phosphorylated (active) form of β-catenin (S33/S37/T41), it is suitable for detecting stabilized β-catenin upon Wnt pathway activation. In WB, a band at approximately 85 kDa is expected; consider validating specificity using knockdown or knockout control samples.
- For IHC-P, it is advisable to optimize antigen retrieval conditions and carefully interpret staining patterns, given the broad subcellular localizations of β-catenin.
- For ELISA, this antibody can be used for detection or capture; however, appropriate controls should be included to ensure signal specificity for active β-catenin.
