Biological Background: Catenin Beta-1 (β-Catenin) Function and Localization

• Serves as a key downstream component of the canonical Wnt signaling pathway. In the absence of Wnt, β-catenin is phosphorylated by a destruction complex (AXIN1, APC, GSK3B, CSNK1A1) and targeted for proteasomal degradation. Upon Wnt stimulation, β-catenin is stabilized, accumulates in the nucleus, and functions as a coactivator for TCF/LEF transcription factors to activate Wnt-responsive genes.
