Biological Background: NOLC1 Function and Localization
- NOLC1 (Nucleolar and coiled-body phosphoprotein 1), also known as Nopp140, p130, NOPP130, NOPP140, or NS5ATP13, is encoded by the NOLC1 gene and undergoes alternative splicing.
- Functions as a regulator of RNA polymerase I by linking RNA polymerase I with enzymes responsible for ribosomal RNA processing and modification. Related references: PMID:10567578, PMID:26399832
- Plays a critical role in neural crest specification: upon monoubiquitination by the BCR(KBTBD8) complex, NOLC1 associates with TCOF1, serving as a platform to connect RNA polymerase I with the ribosomal processing machinery, thereby remodeling the translational program to favor neural crest differentiation. Related references: PMID:26399832
- Involved in nucleologenesis by maintaining the structural integrity of the fibrillar center and dense fibrillar component within the nucleolus; exhibits intrinsic GTPase and ATPase activities. Related references: PMID:9016786
- Localizes predominantly to the nucleolus, but is also found in the nucleus and cytoplasm.
- Subject to multiple post-translational modifications, including acetylation, methylation, phosphorylation, and ubiquitination; contains ATP- and GTP-binding domains.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-100) of human NOLC1; consider this when selecting detection strategies.
- Suitable for Western blot (WB), immunohistochemistry-paraffin (IHC-P), immunofluorescence/immunocytochemistry (IF/ICC), and ELISA. Validate in your specific sample system and optimize antibody dilutions.
- Cross-reactivity is confirmed for human and mouse; potential reactivity with other species should be verified experimentally.
- For nucleolar targets, ensure proper nuclear permeabilization and include appropriate controls to confirm localization.
