Biological Background: NOGO Receptor 1 Function and Localization

  • Reticulon-4 receptor (RTN4R, also known as Nogo receptor) is a GPI-anchored glycoprotein that serves as a receptor for myelin-associated inhibitors, including Nogo (RTN4), OMG, and MAG, thereby mediating axonal growth inhibition and limiting regeneration in the adult CNS.   • RTN4R also binds sialylated gangliosides GT1b and GM1, and by similarity, chondroitin sulfate proteoglycans and heparin, broadening its ligand repertoire. Related references: PMID:18411262

For Western blot, the predicted molecular weight is ~51 kDa; we recommend using brain tissue or neuronal cell lysates as positive controls.
  • For immunofluorescence (IF/ICC), optimize fixation and permeabilization conditions to preserve membrane-associated RTN4R localization. Co-staining with dendritic or axonal markers may aid interpretation.
  • The antibody has demonstrated reactivity with human, mouse, and rat samples; however, we advise validating the antibody in your specific model system, preferably using knockdown or knockout controls, to ensure specificity.
