Biological Background: NOD2 Function and Localization

  • Nucleotide-binding oligomerization domain-containing protein 2 (NOD2, also known as CARD15) is a pattern recognition receptor that detects bacterial peptidoglycan fragments, specifically muramyl dipeptide (MDP) and its phosphorylated form 6-O-phospho-MDP, playing a central role in gastrointestinal immunity. Related references: PMID:12514169 PMID:12527755 PMID:12626759 PMID:36002575
  • Upon MDP binding, NOD2 oligomerizes and recruits the adapter protein RIPK2, leading to NF-kappa-B and MAP kinase pathway activation and transcription of immune response genes. Related references: PMID:11087742 PMID:17355968 PMID:21887730 PMID:15198989
  • NOD2 also functions in antiviral immunity by sensing single-stranded RNA from viruses, leading to IRF3 activation and type I interferon expression via interaction with MAVS. Related references: PMID:19701189
  • In dendritic cells, NOD2 regulates autophagy by recruiting ATG16L1 to sites of bacterial entry, a mechanism that also supports intestinal stem cell survival through mitophagy. Related references: PMID:20637199
  • NOD2 is involved in the endoplasmic reticulum stress response by sensing sphingosine-1-phosphate, which triggers inflammation via NF-kappa-B and MAP kinase signaling. Related references: PMID:27007849 PMID:33942347
  • This protein can activate CASP1 and IL1B release in macrophages through NLRP1 activation following MDP stimulation. Related references: PMID:18511561
  • NOD2 localizes to the basolateral cell membrane, cytoplasm, and mitochondria, and is highly expressed in monocytes, macrophages, dendritic cells, hepatocytes, and intestinal Paneth cells.
  • Mutations in NOD2 are associated with inflammatory bowel diseases such as Crohn's disease and Blau syndrome, highlighting its critical role in immune homeostasis.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 30–216, which encompass the N-terminal CARD domains; therefore the antibody likely recognizes both full-length NOD2 and any N-terminal fragments.
  • For Western blot, the predicted molecular weight is ~115 kDa, but alternative initiation isoforms or post-translational modifications may produce additional bands. Validate in the relevant sample system and consider using appropriate controls.
  • In immunohistochemistry and immunocytochemistry, optimize permeabilization and fixation conditions as NOD2 can be present at the membrane or in the cytoplasm.
  • For ELISA applications, confirm the antibody's performance with the appropriate recombinant protein or peptide standard.
  • When using this antibody in mouse or rat tissues, cross-reactivity is expected, but independent validation is recommended due to species-specific epitope variations.
