Biological Background: NOD1 Function and Localization
- NOD1 (CARD4) is a pattern recognition receptor (PRR) belonging to the NOD-like receptor family, playing a crucial role in innate immunity by detecting bacterial peptidoglycan fragments and other danger signals.
- Specifically recognizes and binds gamma-D-glutamyl-meso-diaminopimelic acid (iE-DAP), a dipeptide present in Gram-negative bacterial peptidoglycan, and preferentially binds tripeptide-containing muropeptides (MurNAc-TriDAP/TriDAP).

- For Western blot, a major band at approximately 108 kDa is expected. Use NOD1-positive lysates (e.g., THP-1, HeLa) as positive controls, and optimize protein loading and transfer conditions.
- For immunohistochemistry (IHC-P), employ standard antigen retrieval methods (e.g., citrate buffer, pH 6.0) and titrate the antibody to achieve optimal signal-to-noise ratio.
- Cross-reactivity with mouse and rat NOD1 is confirmed; this enables testing in common rodent models. Perform in-house verification for your specific tissue or cell type.
- In ELISA, use a direct or capture format and include appropriate positive and negative controls; the antibody may be used as a detection reagent when paired with a suitable capture partner.
