Biological Background: NNMT Function and Localization
- NNMT is a cytoplasmic methyltransferase that catalyzes the N-methylation of nicotinamide, using S-adenosyl-L-methionine (SAM) as a methyl donor, to produce N1-methylnicotinamide (MNA) and S-adenosyl-L-homocysteine (SAH). - By consuming SAM, NNMT reduces the cellular methylation potential, directly limiting SAM availability for other methyltransferases. This activity actively mediates genome-wide epigenetic reprogramming, notably through hypomethylation of repressive histone marks such as H3K27me3. - In early development, NNMT contributes to the low levels of repressive histone marks that characterize the pluripotent pre-implantation state of embryonic stem cells. Related references: PMID:26571212
- NNMT functions as a metabolic regulator in white adipose tissue, influencing energy expenditure, hepatic gluconeogenesis, and cholesterol biosynthesis. In white adipocytes, it regulates polyamine flux and, by consuming nicotinamide, controls NAD+ levels through the salvage pathway.
- The product N1-methylnicotinamide further regulates protein acetylation in hepatocytes by repressing ubiquitination and increasing the stability of SIRT1 deacetylase.

- Subcellular localization: NNMT is predominantly cytoplasmic.
- Tissue expression: NNMT is predominantly expressed in the liver, with lower levels in kidney, lung, skeletal muscle, placenta, and heart. It is not detected in brain or pancreas.
