Biological Background: NNMT Function and Localization
- Nicotinamide N-methyltransferase (NNMT) is a cytoplasmic enzyme encoded by the NNMT gene, playing a key role in vitamin B3 metabolism.
- Catalyzes the N-methylation of nicotinamide using S-adenosyl-L-methionine to form N1-methylnicotinamide, a predominant nicotinamide/vitamin B3 clearance pathway. Related references: PMID:21823666, PMID:23455543, PMID:8182091
- Regulates cellular methylation potential by consuming S-adenosyl-L-methionine, thereby limiting its availability for other methyltransferases and mediating genome-wide epigenetic changes such as H3K27me3 hypomethylation. Related references: PMID:23455543, PMID:26571212, PMID:31043742
- In embryonic stem cells, contributes to the low repressive histone marks characteristic of the pluripotent pre-implantation state. Related references: PMID:26571212
- Acts as a metabolic regulator in white adipose tissue (controlling polyamine flux and NAD+ levels) and in liver (modulating gluconeogenesis and cholesterol biosynthesis) (By similarity).
- Can N-methylate other pyridines structurally related to nicotinamide, suggesting a role in xenobiotic detoxification. Related references: PMID:30044909
- Subcellular localization: Cytoplasm.
- Predominantly expressed in the liver, with lower levels in kidney, lung, skeletal muscle, placenta, and heart; not detected in brain or pancreas.
Experimental Guidance and Technical Tips
- The immunogen spans the full-length human NNMT protein (amino acids 1–264), supporting recognition of native and recombinant forms in multiple applications.
- For Western blot (WB), consider using a loading control and verifying a band at the expected molecular weight of ~30 kDa.
- In immunofluorescence (IF/ICC), cytoplasmic staining is anticipated; optimize fixation and permeabilization conditions for your cell type.
- For immunoprecipitation (IP), titrate the antibody amount relative to lysate concentration to achieve efficient pull-down.
- In ELISA assays, ensure appropriate coating buffers and detection reagents are used; pairing with a suitable secondary antibody is recommended.
- The KD validation tag indicates that this antibody has been tested in knockdown experiments; however, users should validate in their own experimental systems.
