Biological Background: NNMT Function and Localization
- NNMT catalyzes the N-methylation of nicotinamide using S-adenosyl-L-methionine, forming N1-methylnicotinamide and S-adenosyl-L-homocysteine. This reaction represents a predominant nicotinamide/vitamin B3 clearance pathway. Related references: PMID:21823666 PMID:23455543 PMID:8182091
- By consuming S-adenosyl-L-methionine, NNMT regulates cellular methylation potential and mediates genome-wide epigenetic changes, including hypomethylation of repressive histone marks such as H3K27me3. Related references: PMID:23455543 PMID:26571212 PMID:31043742
- NNMT contributes to maintaining low levels of repressive histone marks that characterize pluripotent embryonic stem cell pre-implantation state. Related references: PMID:26571212
- In metabolic tissues, NNMT acts as a regulator of white adipose tissue energy expenditure, hepatic gluconeogenesis, and cholesterol biosynthesis. It also influences polyamine flux and NAD+ levels in adipocytes, and its product N1-methylnicotinamide stabilizes SIRT1 deacetylase in hepatocytes.
- Predominantly localized to the cytoplasm.
- Predominantly expressed in the liver, with lower expression in kidney, lung, skeletal muscle, placenta, and heart. Not detected in brain or pancreas.
- NNMT is subject to post-translational modifications including acetylation and citrullination, and belongs to the S-adenosyl-L-methionine-dependent methyltransferase family.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein covering full-length human NNMT (amino acids 1–264). This design may support recognition of both native and denatured NNMT in assays.
- Validated applications are western blot (WB) and ELISA. For WB, consider using reducing conditions and a 29 kDa expected band. For ELISA, optimize antibody pairing with appropriate detection reagents.
- Given NNMT's high expression in liver, hepatocyte-derived cell lines or liver tissue lysates may serve as suitable positive controls for antibody testing.
- This antibody is specific to human NNMT; cross-reactivity with other species has not been evaluated.
