Biological Background: NME2/NM23B Function and Localization

  • NME2 (Nucleoside diphosphate kinase B), also known as NM23-H2, PUF, or NDKB, catalyzes the transfer of gamma-phosphate from ATP to nucleoside diphosphates via a phosphohistidine intermediate, playing a central role in nucleoside triphosphate homeostasis. Related references: PMID:11121025, PMID:16313181, PMID:1851158
  • It acts as a histidine protein kinase, phosphorylating key signaling proteins like the intermediate-conductance calcium-activated potassium channel KCNN4 at His-358, thereby activating Ca(2+) influx and promoting B and T cell activation. Related references: PMID:17157250, PMID:20946858
  • Beyond its enzymatic activity, NME2 functions as a transcriptional regulator through direct DNA binding and chromatin remodeling. It specifically binds and stabilizes G-quadruplex (G4) structures in the MYC gene promoter, facilitating recruitment of single-strand DNA binding proteins and activating MYC transcription. Related references: PMID:11121025, PMID:11694515, PMID:19033359, PMID:19435876, PMID:25679041
  • Together with NME1, NME2 may regulate acetyl-CoA usage between histone acetylation and fatty acid synthesis by targeting acetyl-CoA release at HAT-associated chromatin regions.
  • NME2 negatively regulates Rho activity by interacting with AKAP13/LBC, providing a link to cytoskeletal dynamics. Related references: PMID:15249197
  • Subcellular localization encompasses cytoplasm, nucleus, lamellipodia, ruffles, and perinuclear region, consistent with its multitasking roles in metabolism, signaling, and transcription.

Experimental Guidance and Technical Tips

  • The immunogen (amino acids 50–152) lies within a conserved region shared across human, mouse, and rat, supporting multi-species applications. Cross-reactivity has been confirmed for mouse and rat samples.
  • For Western blotting, the predicted molecular weight is 17 kDa (17298 Da); use reduced samples and a suitable positive control such as HeLa or Jurkat whole-cell lysate.
  • In ELISA, this monoclonal antibody can serve as either capture or detection reagent; optimization of coating buffer, blocker, and incubation conditions is recommended for individual assay formats.
