Biological Background: NME1/NM23A Function and Localization
- Nucleoside diphosphate kinase A (NDK A/NM23-H1) is a member of the nucleoside diphosphate kinase family, encoded by the NME1 gene, also known as NDPKA, NM23, and GAAD. It is ubiquitously expressed in human tissues and conserved across species.
- Core nucleoside triphosphate homeostasis: Catalyzes the transfer of a gamma-phosphoryl group from ATP to nucleoside diphosphates via a phosphohistidine intermediate, maintaining cellular NTP pools. Also phosphorylates geranyl pyrophosphate (GPP) and farnesyl pyrophosphate (FPP), linking to isoprenoid metabolism. Related references: PMID:10952986, PMID:14960567, PMID:16313181
- Additional kinase activities: Functions as a non-specific serine/threonine kinase and histidine protein kinase, transferring phosphoryl groups to target proteins. Related references: PMID:8529641, PMID:9038158
- Role in DNA fragmentation and apoptosis: As part of the SET complex, NME1 acts as a Mg²⁺-dependent single-stranded DNA endonuclease, cooperating with TREX1 to mediate apoptotic DNA fragmentation in cytotoxic T lymphocytes. It nicks one DNA strand, enabling TREX1 to process the free 3' end, enhancing DNA damage. May also function as a 3'-5' DNA exonuclease. Related references: PMID:12628186, PMID:16818237, PMID:14960567, PMID:16313181
- Tumor metastasis and cell motility: Involved in the regulation of tumor metastasis and cellular differentiation, and is required for cell motility. May control, with NME2, acetyl-CoA usage between histone acetylation and fatty acid synthesis at transcriptionally active chromatin regions. Related references: PMID:8270257, PMID:25582197
- Subcellular localization: Found in the cytoplasm, nucleus, and cell membrane, consistent with its diverse functions in signaling, nucleic acid metabolism, and membrane-associated processes.
- Post-translational modifications and keywords: The protein undergoes acetylation, phosphorylation, glutathionylation, and can form disulfide bonds. Keywords highlight its roles in kinase activity, nucleotide metabolism, apoptosis, and tumor suppression.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 1-100 of human NME1 (P15531). The resulting rabbit monoclonal antibody targets the N-terminal region, which is highly conserved across human, mouse, and rat.
- For Western blotting, consider using whole-cell lysates from human, mouse, or rat cell lines; the expected band weight is approximately 17 kDa under reducing conditions.
- For ELISA, the antibody can be used for direct binding assays with recombinant NME1 protein or the immunogen peptide; appropriate optimization of coating and detection conditions is recommended.
- While cross-reactivity with mouse and rat has been confirmed by the supplier, it is advisable to validate performance in your specific sample matrix and experimental system.
