Biological Background: NLRX1 Function and Localization
- NLRX1, also known as NOD5, NOD9, and CLR11.3, is a member of the NLR family of innate immune receptors that localize to the mitochondria.
- Acts as a negative regulator of MAVS-mediated antiviral responses by inhibiting the virus-induced RLH–MAVS interaction. Related references: PMID:18200010
- Promotes autophagy through interaction with TUFM, recruiting ATG5 and ATG12 to the autophagy machinery. Related references: PMID:22749352
- Regulates MAVS-dependent NLRP3 inflammasome activation, leading to attenuation of apoptosis. Related references: PMID:27393910
- Enhances NF-kappa-B and JUN N-terminal kinase signaling by generating reactive oxygen species. Related references: PMID:18219313
- Exhibits sex-dependent regulation of viral-mediated inflammation and energy metabolism, contributing to sex differences observed in infectious and inflammatory diseases (By similarity).
- Localized to the mitochondrial outer membrane, where it interacts with key antiviral and metabolic pathways.
- Ubiquitously expressed, with strongest expression in mammary gland, heart, and muscle tissues; detected in HeLa, HEK293T, THP-1, HL-60, Raji, and Jurkat cell lines.
Experimental Guidance and Technical Tips
- The immunogen (aa 87-360) covers a segment of the N-terminal region including part of the NACHT domain, which may promote epitope accessibility in native conditions.
- For western blot, expect a specific band at approximately 108 kDa; additional bands from splice variants or post‑translational modifications may appear. Use HeLa or THP-1 lysates as positive controls.
- KO validation ensures high specificity; include NLRX1 knockout cell lysates as negative controls in your experiments.
- Cross-reactivity with mouse and rat enables applications in rodent models; validate expression patterns in relevant tissues.
- For ELISA, consider pairing with an antibody targeting a distinct epitope to develop sandwich immunoassays.
