Biological Background: NLRP3 Function and Localization
- NLRP3 (NACHT, LRR and PYD domains-containing protein 3), also known as cryopyrin, is a cytosolic pattern recognition receptor that serves as the sensor component of the NLRP3 inflammasome. Related references: PMID:16407889, PMID:18403674, PMID:18604214
- Upon activation by pathogens, crystals, or environmental particles, NLRP3 oligomerizes and recruits ASC and pro-caspase-1, forming an inflammasome complex that promotes caspase-1 activation; this leads to cleavage and secretion of pro-inflammatory cytokines IL-1β and IL-18, as well as gasdermin D-mediated pyroptosis.
