Biological Background: NLRP3 Function and Localization

  • NLRP3 (NACHT, LRR and PYD domains-containing protein 3), also known as cryopyrin or CIAS1, is a pivotal sensor component of the NLRP3 inflammasome, a multiprotein complex central to innate immune defense.
  • Upon sensing diverse pathogen- and damage-associated signals—including extracellular ATP, nigericin, crystals of monosodium urate, or amyloid-beta—NLRP3 initiates inflammasome assembly by recruiting PYCARD/ASC and pro-caspase-1, leading to caspase-1 activation, IL-1β/IL-18 secretion, and pyroptotic cell death. Related references: PMID:16407889 PMID:18403674 PMID:18604214 PMID:27432880 PMID:23582325 PMID:28847925 PMID:31189953
  • NLRP3 activation is rigorously controlled; it requires K+ efflux and involves dynein-mediated transport to the microtubule organizing center (MTOC), where it is unlocked by NEK7 before relocalizing to dispersed trans-Golgi network (dTGN) membranes via phosphatidylinositol 4-phosphate binding to form the active inflammasome. Related references: PMID:36442502 PMID:39173637 PMID:30487600 PMID:34554188
  • Beyond its canonical inflammasome role, NLRP3 also functions independently in the nucleus as a transcriptional regulator, promoting Th2 cell differentiation by binding to DNA and driving IL4 expression.
  • The protein dynamically localizes to multiple subcellular compartments, including the cytosol, mitochondria, Golgi apparatus, endoplasmic reticulum, and nucleus, and can be secreted.
  • Predominantly expressed in macrophages, NLRP3 is also present in dendritic cells, B- and T-cells, as well as in stratified squamous epithelia and thymic Hassall's corpuscles. Related references: PMID:33231615 PMID:34133077 PMID:11786556 PMID:17164409
  • Key post-translational modifications—including phosphorylation, ADP-ribosylation, palmitoylation, disulfide bond formation, and ubiquitination—finely tune NLRP3 stability, localization, and activity.

Experimental Guidance and Technical Tips

  • For Western blotting, consider using a low-percentage SDS-PAGE gel (e.g., 8%) and optimizing transfer conditions for high-molecular-weight NLRP3 (~118 kDa). A positive control such as LPS-stimulated THP-1 macrophage lysate may be beneficial.
  • This rabbit monoclonal antibody is suitable for standard ELISA formats; when setting up your assay, evaluate coating concentrations and pair with an appropriate detection antibody in your specific sample matrix.
  • Given that NLRP3 expression can be induced under inflammatory conditions, pre-treatment of cell models with NLRP3 activators (e.g., LPS plus nigericin) may enhance detection in both WB and ELISA.
