Biological Background: NLRP3 Function and Localization
- NLRP3 (NACHT, LRR and PYD domains-containing protein 3), also known as cryopyrin or CIAS1, is a pivotal sensor component of the NLRP3 inflammasome, a multiprotein complex central to innate immune defense.
- Upon sensing diverse pathogen- and damage-associated signals—including extracellular ATP, nigericin, crystals of monosodium urate, or amyloid-beta—NLRP3 initiates inflammasome assembly by recruiting PYCARD/ASC and pro-caspase-1, leading to caspase-1 activation, IL-1β/IL-18 secretion, and pyroptotic cell death.
