- NLRP2 is a member of the NOD-like receptor (NLR) family, characterized by an N-terminal pyrin domain (PYD), a central NACHT domain with ATPase activity, and C-terminal leucine-rich repeats (LRRs).
- It functions as a negative regulator of NF-kB signaling by suppressing TNF- and CD40-induced NFKB1 activity at the IKK complex level, inhibiting NFKBIA degradation.
- Upon association with the adaptor protein PYCARD (ASC), NLRP2 activates caspase-1 (CASP1), promoting the maturation and secretion of the pro-inflammatory cytokine IL-1β, likely as part of an inflammasome complex.
- The protein is expressed at high levels in lung, placenta, and thymus, and at lower levels in ovary, intestine, and brain. It is also highly abundant in oocytes and early embryos.
