Biological Background: NLRP12 Function and Localization

  • NLRP12 is also known as Monarch-1, PYPAF7, and RNO, encoded by the NLRP12 gene in humans.
  • It functions as a potent negative regulator of inflammation, inhibiting both canonical and non-canonical NF-κB and ERK signaling pathways in dendritic cells and macrophages. Related references: PMID:15489334, PMID:17947705
  • NLRP12 promotes bacterial tolerance by targeting NOD2 for proteasomal degradation. Related references: PMID:30559449
  • In antiviral immunity, it suppresses RIG-I-mediated signaling by modulating ubiquitination of RIG-I, dampening type I interferon responses. Related references: PMID:30902577
  • NLRP12 is predominantly expressed in peripheral blood leukocytes, especially eosinophils and granulocytes, with lower levels in monocytes.
  • The protein localizes to the cytoplasm and contains leucine-rich repeats, a nucleotide-binding domain, and a PYRIN domain.
  • Structurally, NLRP12 belongs to the NLR family, featuring an N-terminal PYRIN domain, a central NACHT domain, and C-terminal leucine-rich repeats.
  • It has been implicated in mitigating obesity-related inflammation in adipose tissue (By similarity).

Experimental Guidance and Technical Tips

  • For Western blotting, consider using appropriate positive controls such as human leukocyte lysates and verify the expected band size (~120 kDa).
  • Optimize antibody dilution and blocking conditions for WB; inclusion of loading controls and molecular weight markers is essential.
  • ELISA can be used for antibody titer determination or target detection; validate with recombinant NLRP12 protein or known positive samples.
  • Ensure cross-reactivity validation in human-derived samples; note that reactivity in other species has not been tested.
  • The immunogen corresponds to the C-terminal region (aa 862-1061); confirm epitope specificity if detecting isoforms.
