Biological Background: NLRP12 Function and Localization
- NLRP12 is a potent mitigator of inflammation, primarily expressed in dendritic cells and macrophages, where it inhibits both canonical and non-canonical NF-κB and ERK activation pathways. Related references: PMID:15489334, PMID:17947705, PMID:30559449
- It functions as a negative regulator of NOD2 by targeting it for proteasomal degradation, thereby promoting bacterial tolerance. Related references: PMID:30559449
- NLRP12 also inhibits RIGI-mediated immune signaling against RNA viruses by reducing TRIM25-mediated 'Lys-63'-linked RIGI activation and enhancing RNF125-mediated 'Lys-48'-linked RIGI degradation. Related references: PMID:30902577
- It acts as a negative regulator of inflammatory response in adipose tissue, helping to mitigate obesity and obesity-associated diseases.
- Subcellularly, NLRP12 is localized in the cytoplasm.
- NLRP12 is predominantly expressed in peripheral blood leukocytes, especially eosinophils and granulocytes, with lower levels in monocytes.
- The protein contains key structural domains including a pyrin domain (PYD), a NACHT domain with ATP-binding, and C-terminal leucine-rich repeats (LRRs).
- Alternative splicing of NLRP12 produces multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 862–1061) targets the C-terminal region of human NLRP12, which encompasses the leucine-rich repeat (LRR) domain; confirm that the antibody recognizes full-length NLRP12 in your model system.
- For western blotting, expect a band at approximately 120 kDa; use a positive control such as peripheral blood leukocyte lysates or NLRP12-overexpressing cells, and validate with siRNA knockdown or knockout lysates when possible.
- This antibody is also applicable for ELISA; determine optimal coating concentrations and dilution factors using purified recombinant NLRP12 protein.
- Noted cross-reactivity with rat; verify species reactivity in rat-derived samples using appropriate positive and negative controls.
- As a polyclonal antibody, evaluate lot-to-lot consistency if used in long-term studies or diagnostic assay development.
