- Protein identity: Homeobox protein Nkx-3.1 (NKX3-1, also designated Homeobox protein NK-3 homolog A) is a transcription factor encoded by the NKX3-1 gene and characterized by a homeobox DNA-binding domain. Alternative splicing produces multiple isoforms.
- DNA-binding and transcription regulation: NKX3.1 preferentially binds the consensus sequence 5'-TAAGT[AG]-3' and functions primarily as a transcriptional repressor, modulating gene expression programs in target cells.
- Prostate development: It plays a critical role in normal prostate development, controlling proliferation of the glandular epithelium and directing proper ductal morphogenesis.
- Tumor suppressor: NKX3.1 acts as a tumor suppressor in prostate carcinogenesis; it has been shown to inhibit proliferation and invasion of PC-3 prostate cancer cells.
- Subcellular localization: The protein is predominantly localized to the nucleus, consistent with its role in transcriptional regulation.
- Tissue specificity: NKX3.1 is highly expressed in the prostate, with lower levels detectable in the testis.
- Post-translational modifications and molecular features: The protein is subject to ubiquitin conjugation (Ubl conjugation) and has been identified in proteomics screens. It contains a homeobox domain essential for DNA binding and transcriptional activity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-100) of human NKX3.1; consider this when analyzing isoforms or truncated forms.
- For Western blot (WB), a band around 26 kDa is expected. Verify specificity using lysates from prostate or prostate cancer cell lines (e.g., LNCaP, PC-3) as positive controls, and include negative controls from tissues with low or absent expression.
- For immunohistochemistry (IHC-P), nuclear staining should be observed. Antigen retrieval and antibody dilution may require optimization depending on the tissue fixation and processing conditions.
- Due to reactivity across human, mouse, and rat, the antibody is suitable for cross-species studies, but validation in each species’ sample system is recommended.
