- NKX2-1 (Homeobox protein Nkx-2.1), also known as thyroid transcription factor 1 (TTF-1), is a member of the NK-2 homeobox family of transcription factors.
- It plays a critical role in thyroid development and function by activating thyroid-specific genes such as thyroglobulin, thyroperoxidase, and thyrotropin receptor.
- Involved in lung development and surfactant homeostasis; forms a regulatory loop with GRHL2 that coordinates lung epithelial cell morphogenesis and differentiation.
- Activates the transcription of GNRHR and enhances the circadian oscillation of its gene expression, while repressing the circadian transcriptional repressor NR1D1 (by similarity).
- Localizes to the nucleus, consistent with its function as a DNA-binding transcription factor.
- Undergoes alternative splicing and is a phosphoprotein, suggesting regulation by post-translational modifications.
