Biological Background: NKX2-1 Function and Localization

  • NKX2-1 (Homeobox protein Nkx-2.1), also known as thyroid transcription factor 1 (TTF-1), is a member of the NK-2 homeobox family of transcription factors.
  • It plays a critical role in thyroid development and function by activating thyroid-specific genes such as thyroglobulin, thyroperoxidase, and thyrotropin receptor.
  • Involved in lung development and surfactant homeostasis; forms a regulatory loop with GRHL2 that coordinates lung epithelial cell morphogenesis and differentiation.
  • Activates the transcription of GNRHR and enhances the circadian oscillation of its gene expression, while repressing the circadian transcriptional repressor NR1D1 (by similarity).
  • Localizes to the nucleus, consistent with its function as a DNA-binding transcription factor.
  • Undergoes alternative splicing and is a phosphoprotein, suggesting regulation by post-translational modifications.
  • Tissue-specific expression is predominantly in thyroid and lung.

Experimental Guidance and Technical Tips

  • The immunogen was designed from the N-terminal region (aa 20–110) of human NKX2-1. For Western blot, consider enriching nuclear extracts from thyroid or lung tissue/cell lines to improve detection.
  • Given the reported cross-reactivity with mouse and rat, validate the antibody in your specific sample system, as epitope conservation may vary.
  • For ELISA applications, ascertain whether the antibody recognizes the denatured or native form of the protein as required by your assay format.
  • Because NKX2-1 may undergo alternative splicing and phosphorylation, the observed band size may differ slightly from the predicted 39 kDa; confirm with appropriate controls.
