Biological Background: Ninjurin-1 Function and Localization

• Ninjurin-1 (Nerve injury-induced protein 1) is a small transmembrane protein (16.5 kDa) encoded by the Ninj1 gene in mouse. It belongs to the ninjurin family and is also known as NIN1 or NINJURIN.

• NINJ1 functions as a key effector of programmed cell death, including pyroptosis and necroptosis. It mediates plasma membrane rupture (cytolysis) downstream of gasdermin family members or MLKL, leading to release of damage-associated molecular patterns (DAMPs) that propagate inflammation. • Upon activation, NINJ1 oligomerizes and forms ring-like structures on the plasma membrane.
