Biological Background: Ninein Function and Localization
- Ninein is a centrosomal protein encoded by the NIN gene, also known as glycogen synthase kinase 3 beta-interacting protein (GSK3B-interacting protein).
- Ninein is required for positioning and anchorage of the microtubule minus-end in epithelial cells. Related references: PMID:15190203 PMID:23386061
- It acts as a centrosome maturation factor. Related references: PMID:11956314
- Ninein plays a role in microtubule nucleation by recruiting the gamma-tubulin ring complex to the centrosome. Related references: PMID:15190203
- Overexpression of ninein suppresses release of microtubules without perturbing nucleation or elongation. Related references: PMID:15190203
- Required for centriole organization and microtubule anchoring at the mother centriole. Related references: PMID:23386061
- Subcellular localization includes cytoplasm, centrosome, centriole, cell junctions, and desmosomes.
- Ubiquitously expressed with highest levels in heart and skeletal muscle. Isoform 1 is more abundant than isoform 5.
- Contains coiled-coil domains, GTP-binding motifs, and is subject to alternative splicing and phosphorylation. Associated with disease variants (dwarfism, intellectual disability).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-287), so this antibody may recognize multiple isoforms sharing this region. Consider validating with isoform-specific controls.
- For Western blotting, the target protein weight is ~243 kDa. Use low-percentage gels and extended transfer times to ensure efficient detection.
- In IHC-P, perform antigen retrieval and optimize blocking steps. Positive control tissues with high ninein expression (e.g., heart, skeletal muscle) are recommended.
- Cross-reactivity with mouse and rat suggests epitope conservation. Validate reactivity in these species with appropriate positive and negative controls.
- For ELISA, determine optimal coating concentration and antibody dilution experimentally to achieve desired sensitivity and specificity.
