Biological Background: NHLH1 Function and Localization
- NHLH1 (also known as HEN1, NSCL, bHLHa35) is a class A basic helix-loop-helix (bHLH) transcription factor encoded by the NHLH1 gene on human chromosome 1.
- The protein contains a conserved bHLH domain that is predicted to mediate sequence-specific DNA binding and protein dimerization, characteristic of transcriptional regulators.
- NHLH1 is localized to the nucleus, consistent with its role in transcriptional regulation and cell fate determination.
- Functional studies suggest that NHLH1 may serve as a DNA-binding protein involved in the control of cell-type determination, possibly within the developing nervous system.
- It is annotated as a developmental protein and differentiation factor, indicating a role in early embryonic development and neurogenesis.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal region (amino acids 1–60), which lies outside the conserved bHLH domain. Consider potential cross-reactivity with other bHLH family members if epitope similarity exists.
- For Western blotting, the predicted molecular weight is approximately 15 kDa. However, post-translational modifications, dimerization, or alternative translation start sites may result in bands at higher molecular weights; validate using appropriate controls.
- Due to its nuclear localization, this antibody may be suitable for chromatin immunoprecipitation (ChIP) assays. Optimize cross-linking and chromatin shearing conditions for your specific cell type or tissue.
- For immunoprecipitation, use non-denaturing lysis buffers to maintain protein–protein interactions, and include appropriate IgG controls to assess specificity.
- Cross-reactivity data is currently available only for human samples. If using in other species, verify sequence homology of the immunogen region and validate experimentally.
