Biological Background: NHEJ1 Function and Localization
- NHEJ1, also known as XLF or Cernunnos, is a DNA repair protein essential for non-homologous end joining (NHEJ), double-strand break repair, and V(D)J recombination. Related references: PMID:16439204, PMID:16439205, PMID:17317666
- It promotes the ligation of various mismatched and non-cohesive DNA ends, facilitating efficient repair. Related references: PMID:17470781, PMID:17717001, PMID:19056826
- Together with PAXX, NHEJ1 collaborates with DNA polymerase lambda (POLL) to promote joining of non-cohesive DNA ends. Related references: PMID:25670504, PMID:30250067
- NHEJ1 interacts with XRCC4 to form filaments or bridges that hold broken DNA ends in close proximity, stimulating ligation by the XRCC4-DNA ligase IV complex. Related references: PMID:21768349, PMID:21775435, PMID:22228831, PMID:27437582, PMID:28500754
- It binds DNA in a length-dependent manner, with preference for longer DNA substrates. Related references: PMID:17317666, PMID:18158905
- NHEJ1 is localized to the nucleus and chromosomes, consistent with its role in genome maintenance.
- It is ubiquitously expressed across human tissues, highlighting its fundamental role in DNA repair.
- Post-translational modifications include phosphorylation, and the protein is subject to alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen comprises full-length human NHEJ1 (amino acids 1-299), suggesting the antibody is suitable for detecting denatured NHEJ1 in Western blot (WB).
- For ELISA applications, consider coating with the recombinant immunogen or NHEJ1 peptides and titrate the antibody to determine optimal working concentrations.
- Validate the antibody’s specificity using NHEJ1 knockout cell lines or siRNA-mediated knockdown to confirm band disappearance in WB.
- Cross-reactivity with mouse NHEJ1 is reported; however, we recommend verifying reactivity in your specific mouse strain or tissue due to potential sequence variations.
- As a polyclonal antibody, lot-to-lot consistency may vary; it is advisable to re-validate each new lot for critical assays.
