Biological Background: NgR3 (RTN4RL1) Function and Localization

  • Reticulon-4 receptor-like 1 (RTN4RL1), also known as Nogo receptor-like 2, Nogo-66 receptor homolog 2, or NgR3, is a GPI-anchored cell surface receptor predominantly expressed in the brain.
  • It plays a functionally redundant role in postnatal brain development and regulating axon regeneration in the adult central nervous system; contributes to normal axon migration across the brain midline and formation of the corpus callosum.
  • NgR3 inhibits neurite outgrowth and axon regeneration by binding to neuronal chondroitin sulfate proteoglycans and heparin, and mediates protection of motoneurons against apoptosis, likely through MAG interaction.
  • It restricts the number of dendritic spines and synapses formed during brain development, signaling through Rho activation and downstream actin cytoskeleton reorganization. Related references: PMID:22325200
  • Subcellular localization includes the cell membrane, membrane rafts, perikaryon, and cell projections, consistent with its receptor function in neurons.

  • For Western blot (WB), a band at approximately 49 kDa is expected. Use lysates from human, mouse, or rat brain tissue as positive controls.
  • When performing immunohistochemistry (IHC-P) or immunofluorescence (IF/ICC), consider antigen retrieval and suitable permeabilization/fixation protocols optimized for neuronal tissues.
  • For ELISA applications, the antibody's reactivity may be assessed against recombinant NgR3 protein or peptide; cross-reactivity with related family members should be evaluated.
