Biological Background: Neuroguidin Function and Localization

  • NGDN (Neuroguidin), also designated CANu1, LCP5, or C14orf120, is an EIF4E-binding protein encoded by the NGDN gene in humans.
  • It is an integral component of the small subunit (SSU) processome, the primary precursor of the small ribosomal subunit. Within the nucleolus, NGDN participates in pre-rRNA processing, facilitating RNA folding, modifications, rearrangements, and cleavage. Its timed dissociation governs the transition from the pre-A1 to pre-A1* assembly state. Related references: PMID:34516797
  • NGDN functions as a translational repressor by inhibiting mRNA translation in a cytoplasmic polyadenylation element (CPE)-dependent manner.
  • Subcellularly, the protein localizes to the nucleus, nucleolus, and centromere, as well as the cytoplasm and cell projections including axons, dendrites, and filopodia.
  • Post-translational modifications include acetylation and phosphorylation, and alternative splicing generates multiple isoforms.
  • The protein features coiled-coil domains and is involved in key processes such as ribosome biogenesis and translation regulation.

Experimental Guidance and Technical Tips

  • The immunogen consists of a recombinant fusion protein spanning amino acids 61–209 of human NGDN; the resulting polyclonal antibody may detect multiple isoforms or post-translationally modified forms.
  • For Western blotting, the predicted molecular weight is 36 kDa; additional bands arising from modifications or cleavage products should be considered and validated in relevant Human, Mouse, or Rat lysates.
  • In ELISA applications, use the recombinant immunogen as a positive control and optimize coating conditions.
  • As a polyclonal reagent, each batch may exhibit slight variations; it is advisable to include appropriate positive and negative controls and verify performance in the intended sample matrix.
