- NG2/CSPG4 is a chondroitin sulfate proteoglycan, also known as melanoma chondroitin sulfate proteoglycan (MCSP), encoded by the CSPG4 gene. It is a large transmembrane protein with an extracellular domain harboring chondroitin sulfate glycosaminoglycan chains.
- Functions as a cell surface receptor for collagen alpha 2(VI) and various growth factors, modulating cell proliferation, migration, and microvascular morphogenesis. It also inhibits neurite outgrowth and growth cone collapse during axon regeneration.
- Plays a critical role in melanoma cell invasion by regulating MPP16-dependent degradation of type I collagen and enhancing plasminogen activation while inhibiting angiostatin, thereby promoting tissue remodeling and angiogenesis.
- Acts as a signal transducer via its cytoplasmic C-terminus, interacting with scaffolding and signaling proteins to promote retraction fiber formation, cell polarization, and integrin-mediated adhesion through Rho GTPase, CDC42, ACK1, and BCAR1 pathways.
- Activates FAK and ERK1/ERK2 signaling cascades, contributing to cell spreading and migration.
- Localizes primarily to the cell membrane, including the apical cell membrane, lamellipodium membrane, and cell surface projections.
