Biological Background: NG2/CSPG4 Function and Localization

  • NG2/CSPG4 is a chondroitin sulfate proteoglycan, also known as melanoma chondroitin sulfate proteoglycan (MCSP), encoded by the CSPG4 gene. It is a large transmembrane protein with an extracellular domain harboring chondroitin sulfate glycosaminoglycan chains.
  • Functions as a cell surface receptor for collagen alpha 2(VI) and various growth factors, modulating cell proliferation, migration, and microvascular morphogenesis. It also inhibits neurite outgrowth and growth cone collapse during axon regeneration.
  • Plays a critical role in melanoma cell invasion by regulating MPP16-dependent degradation of type I collagen and enhancing plasminogen activation while inhibiting angiostatin, thereby promoting tissue remodeling and angiogenesis.
  • Acts as a signal transducer via its cytoplasmic C-terminus, interacting with scaffolding and signaling proteins to promote retraction fiber formation, cell polarization, and integrin-mediated adhesion through Rho GTPase, CDC42, ACK1, and BCAR1 pathways.
  • Activates FAK and ERK1/ERK2 signaling cascades, contributing to cell spreading and migration.
  • Localizes primarily to the cell membrane, including the apical cell membrane, lamellipodium membrane, and cell surface projections.
  • Detected at the protein level in fibroblasts, placenta, and malignant melanoma cells, underscoring its importance in cancer progression and developmental processes.
  • Post-translational modifications include phosphorylation, disulfide bond formation, and N-linked glycosylation. It features a transmembrane helix and a signal peptide, classifying it as a type I transmembrane proteoglycan.

Experimental Guidance and Technical Tips

  • The immunogen (aa 1950–2210) resides in the cytoplasmic domain; therefore, the antibody may detect both full-length and truncated forms of CSPG4 in western blotting.
  • Due to the large molecular weight (~251 kDa), consider using a low-percentage SDS-PAGE gel (e.g., 6%) and extended transfer times for optimal WB results.
  • For IHC-P, heat-induced antigen retrieval may be required to expose the epitope in formalin-fixed, paraffin-embedded tissues.
  • In IF/ICC, permeabilization steps are essential to allow antibody access to the intracellular epitope.
  • The confirmed cross-reactivity with mouse enables comparative studies, but validation in the specific sample system is recommended due to potential species-dependent differences.
  • When designing ELISA assays, pairing this antibody with a capture antibody targeting an extracellular epitope may yield effective sandwich immunoassays.
