Biological Background: NG2/CSPG4 Function and Localization

  • Chondroitin sulfate proteoglycan 4 (NG2/CSPG4), also known as melanoma chondroitin sulfate proteoglycan, is a large integral membrane proteoglycan involved in cell proliferation, migration, and microvascular morphogenesis.
  • It functions as a cell surface receptor for collagen alpha 2(VI), facilitating cellular migration on collagen-rich substrates. NG2/CSPG4 binds growth factors and extracellular matrix proteases, modulating their activity and contributing to melanoma cell invasion through MPP16-dependent type I collagen degradation.
  • Subcellularly, NG2/CSPG4 localizes to the cell membrane, apical membrane, lamellipodium membrane, and cell surface, consistent with roles in cell polarity, adhesion, and migration.
  • Tissue expression is prominent in malignant melanoma cells, and also detected in fibroblasts and placenta at the protein level. Related references: PMID:36213313 PMID:32337544
  • As a signal transducer, the cytoplasmic domain recruits scaffolding and signaling proteins, promoting retraction fiber formation, cell polarization via Rho GTPase, and integrin‑mediated adhesion through CDC42, ACK1, and BCAR1 cascades. It also activates FAK and ERK1/ERK2 pathways.
  • Key structural features include disulfide bonds, a transmembrane domain, and multiple glycosylation/phosphorylation sites, classifying it as a glycoprotein and phosphoprotein involved in angiogenesis, developmental processes, and tissue remodeling.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 1340–1563 near the C‑terminal extracellular region; thus, the antibody is expected to recognize the full‑length ~251 kDa NG2/CSPG4 protein in western blotting. Validate detection in human melanoma cell lines (e.g., A375) or CSPG4‑transfected cells.
  • For ELISA, consider using recombinant CSPG4 extracellular domain fragments or whole cell lysates from CSPG4‑expressing cells. Optimize coating and detection conditions based on the specific assay format.
  • Given the high molecular weight, use a 6–8% SDS‑PAGE gel and extended transfer time for western blot to ensure efficient transfer of the large proteoglycan.
