Biological Background: NG2 (CSPG4) Function and Localization
- NG2, also known as chondroitin sulfate proteoglycan 4 (CSPG4) or melanoma-associated chondroitin sulfate proteoglycan (MCSP), is a large cell-surface proteoglycan implicated in cell proliferation, migration, and angiogenesis.
- It acts as a receptor for collagen alpha-2(VI), facilitating cell migration on collagen-rich substrates, and binds growth factors and extracellular proteases via its N-terminal domain, modulating their activity.
- NG2 inhibits neurite outgrowth and growth cone collapse, suggesting a role in restricting axon regeneration after injury.
- The cytoplasmic C-terminus functions as a signal transduction hub, interacting with scaffolding and signaling proteins to promote retraction fiber formation, cell polarization through Rho GTPase activation, and integrin-mediated adhesion via the CDC42-ACK1-BCAR1 cascade.
- NG2 enhances FAK and ERK1/ERK2 signaling, further supporting its role in cell motility and invasion, particularly in melanoma cells where it may aid type I collagen degradation.
