Biological Background: NFS1 Function and Localization
- NFS1 (also known as IscS, NIFS, or COXPD52) is a pyridoxal phosphate-dependent cysteine desulfurase that releases sulfur from L-cysteine to L-alanine for iron-sulfur (Fe-S) cluster biogenesis. Related references: PMID:18650437 PMID:29097656 PMID:31101807
- NFS1 is the catalytic subunit of the cysteine desulfurase complex (NFS1:LYRM4:NDUFAB1), which generates persulfide sulfur that is transferred to the scaffold protein ISCU, a step assisted by frataxin (FXN).
- The core ISC assembly complex, including NFS1, is critical for the de novo synthesis of [2Fe-2S] clusters, the first step in mitochondrial iron-sulfur protein biogenesis, with subsequent cluster transfer to chaperone proteins HSCB, HSPA9, and GLRX5.

- NFS1 localizes primarily to mitochondria but has been detected in the cytoplasm, nucleus, and centrosome.
- Predominantly expressed in heart and skeletal muscle, with lower levels in brain, liver, and pancreas.
- Mutations in NFS1 are associated with primary mitochondrial disease, and the enzyme requires pyridoxal phosphate as a cofactor.
- Alternative initiation and splicing generate multiple isoforms of NFS1.

- The immunogen sequence (aa 290–457, C-terminal) may detect full-length NFS1 and certain isoforms; consider verifying in the relevant sample system.
- For western blotting, the predicted molecular weight is ~50 kDa, but post-translational modifications or alternative splicing may result in additional bands.
- In IHC-P and IF/ICC, optimize antigen retrieval and fixation conditions, and validate using positive and negative tissue controls.
