Biological Background: NFKBIB Function and Localization

  • NFKBIB encodes NF-kappa-B inhibitor beta (I-kappa-B-beta, IkB-beta), a member of the I-kappa-B family of inhibitors that regulate NF-kappa-B transcription factors.
  • It inhibits NF-kappa-B by complexing with and sequestering it in the cytoplasm, preventing its nuclear translocation and transcriptional activity.
  • Upon cell stimulation, resynthesized unphosphorylated IkB-beta can bind NF-kappa-B and transport it to the nucleus, protecting it from NFKBIA (I-kappa-B-alpha)-mediated inactivation.
  • Interaction with NKIRAS1 and NKIRAS2 prevents IkB-beta phosphorylation, making it more resistant to degradation and explaining its slower turnover compared to I-kappa-B-alpha.
  • The protein localizes to both the cytoplasm and nucleus, consistent with its dynamic role in NF-kappa-B shuttling.
  • It is expressed in all tissues examined, indicating a ubiquitous regulatory function.
  • The protein contains ANK repeats and is subject to phosphorylation; alternative splicing produces multiple isoforms.
  • Synonyms include IKBB, TRIP9, and NFKBIB.

Experimental Guidance and Technical Tips

  • Since the immunogen covers the full-length protein (aa 1-356), the antibody may recognize multiple isoforms or cleavage products; consider validating specificity in your experimental system.
  • For Western blot, a band around 38 kDa is expected; verify against appropriate controls and molecular weight markers.
  • For IHC-P, optimize antigen retrieval and antibody dilution using known positive and negative tissues.
  • Given cross-reactivity with mouse and rat, this antibody is suitable for studies across these species; however, confirm species-specific performance in your assays.
