- NFIL3 (Nuclear factor interleukin-3-regulated protein), also known as E4BP4, IL3BP1, NFIL3A, or NF-IL3A, is a transcriptional regulator encoded by the NFIL3 gene in humans (GeneID: 4783). It recognizes and binds to the sequence 5'-[GA]TTA[CT]GTAA[CT]-3', present in many cellular and viral promoters.
- Acts as both a transcriptional repressor and activator. It represses transcription from promoters with ATF sites and competes with PAR DNA-binding factors (DBP, HLF, TEF) for the same consensus site. It activates transcription from the interleukin-3 promoter in T-cells.
- NFIL3 is a core component of the circadian clock, functioning as a negative regulator of PER2 expression, thereby influencing circadian oscillation.
- Subcellularly localized to the nucleus, consistent with its role as a transcription factor. It is expressed in various tissues including bladder, stomach, thyroid, spinal cord, lymph node, trachea, adrenal gland, bone marrow, and muscle.
- Post-translationally modified by phosphorylation and ubiquitination (isopeptide bond, Ubl conjugation), which may regulate its activity and stability.
- Plays a role in immune cell development; it protects pro-B cells from apoptosis and is required for natural killer cell precursor development.
