Biological Background: NFE2L1 Function and Localization
- NFE2L1 (NRF1/TCF11/LCR-F1) is a CNC-type bZIP transcription factor that functions as an endoplasmic reticulum membrane sensor. It translocates to the nucleus upon cleavage by DDI2 in response to stresses such as cholesterol excess, oxidative stress, or proteasome inhibition. Related references: PMID:20932482, PMID:24448410
- In the nucleus, NFE2L1 heterodimerizes with small Maf proteins (MAFF, MAFG, MAFK) and binds antioxidant response elements (AREs), thereby regulating genes critical for oxidative stress defense and detoxification. Related references: PMID:8932385, PMID:9421508
- It acts as a key sensor of cholesterol excess: at the ER membrane, it directly binds cholesterol via its CRAC motif, blocking its cleavage and activation, thus allowing expression of cholesterol removal genes such as CD36.

When used in WB, a band around 85 kDa is expected for the full-length protein; shorter isoforms or cleavage products may also be observed.
- For IHC-P, consider antigen retrieval protocols (e.g., heat-induced epitope retrieval in citrate buffer, pH 6.0). Optimize antibody concentration based on tissue type and fixation.
- In IF/ICC, ensure appropriate permeabilization to detect nuclear and ER-localized protein. Co-staining with organelle markers may aid interpretation.
