Biological Background: NFATC3 Function and Localization

  • NFATC3 (Nuclear factor of activated T-cells, cytoplasmic 3), also known as NFAT4 or NFATx, is a member of the NFAT family of transcription factors that regulate gene expression in response to calcium signaling.
  • Binds to DNA promoter regions to control transcription of target genes, including TNFSF11/RANKL in osteoblasts and IL-2 in T-cells. Related references: PMID:18815128
  • Translocates from the cytoplasm to the nucleus upon calcium influx via ORAI1 CRAC channels, enabling transcriptional activation of downstream targets.
  • Plays a role in cardiac development and cardiomyocyte survival; forms a complex with STAT3 and NFATC4 to activate CRYAB transcription.
  • Promotes smooth muscle hypertrophy and differentiation by inducing ACTA2 expression in response to EDN1 and ROCK signaling.
  • Predominantly expressed in thymus and skeletal muscle, with weaker expression in kidney, heart, testis, and other tissues. Related references: PMID:9759864
  • Subject to multiple post-translational modifications, including acetylation, phosphorylation, and ubiquitination, which modulate its activity and subcellular localization.
  • Also functions in macrophage-mediated innate immunity by promoting NOS2 transcription and nitric oxide production for pathogen clearance.

  • The immunogen corresponds to the N-terminal region (aa 1–290) of human NFATC3, which may detect multiple isoforms; consider validating isoform specificity in your experimental system.
  • For Western blot (WB), a band near 116 kDa is expected. Validate detection using positive control tissues (e.g., thymus, skeletal muscle) and appropriate loading controls.
  • When performing immunohistochemistry (IHC-P) or immunofluorescence (IF/ICC), include antigen retrieval and proper negative controls to confirm staining specificity.
  • This antibody is compatible with ELISA; determine optimal working dilutions through titration experiments.
  • Cross-reactivity with mouse and rat has been documented; verify reactivity if using samples from other species.
