Biological Background: NFATC3 Function and Localization
- NFATC3 is a member of the nuclear factor of activated T-cells (NFAT) family of transcription factors, also known as NFAT4 or NFATx. It plays a role in Ca(2+)-mediated transcriptional responses upon Ca(2+) influx via ORAI1 CRAC channels.
- Acts as a transcriptional activator by binding to the promoter regions of target genes, including TNFSF11/RANKL and IL-2, thereby promoting their expression. Related references: PMID:18815128
- In T-cells, NFATC3 is critical for the inducible expression of cytokine genes, especially IL-2, contributing to immune response regulation.
- Predominantly expressed in thymus and skeletal muscle, with weaker expression in peripheral blood leukocytes, kidney, testis, spleen, prostate, ovary, small intestine, heart, placenta, and pancreas. Related references: PMID:9759864
- Subcellular localization includes both cytoplasm and nucleus; it shuttles to the nucleus upon activation to regulate gene transcription.
