Biological Background: NFAT5 Function and Localization

  • NFAT5, also known as T-cell transcription factor NFAT5, tonicity-responsive enhancer-binding protein (TonEBP), NFATZ, OREBP, NF-AT5, NFATL1, and TONEBP, is a member of the nuclear factor of activated T-cells family.
  • Functions as a transcription factor involved in the transcriptional regulation of osmoprotective and inflammatory genes, mediating the cellular response to hypertonicity. Related references: PMID:10377394 PMID:11485737 PMID:41652173 PMID:10051678
  • Binds the DNA consensus sequence 5'-[ACT][AG]TGGAAA[CAT]A[TA][ATC][CA][ATG][GT][GAC][CG][CT]-3' to regulate target gene expression. Related references: PMID:10377394 PMID:11485737
  • Positively regulates the transcription of LCN2 and S100A4 genes; optimal transactivation of these genes requires the presence of DDX5/DDX17. Related references: PMID:22266867
  • Involved in the DNA damage response by preventing formation of R-loops, which are composed of a DNA:RNA hybrid and the associated non-template single-stranded DNA. Related references: PMID:34049076
  • Predominantly localizes to the nucleus, but can also be found in the cytoplasm and associated with chromosomes.
  • Widely expressed, with highest levels in skeletal muscle, brain, heart, and peripheral blood leukocytes.
  • Post-translationally modified by acetylation, phosphorylation, ADP-ribosylation, and ubiquitination, which regulate its activity and stability.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal region (aa 1355-1455) of human NFAT5; this sequence may be conserved across isoforms, making the antibody suitable for detecting total NFAT5 protein in Western blot.
  • For Western blot, consider using a low-percentage gel and extended transfer times due to the large molecular weight (~166 kDa) of NFAT5.
  • The antibody reactivity is validated for human NFAT5 by WB and ELISA; cross-reactivity with other species or family members should be tested experimentally.
  • In ELISA applications, the antibody can be considered as a capture or detection reagent after appropriate validation in the relevant sample matrix.
