Biological Background: NFAT2 Function and Localization
- NFAT2, also known as nuclear factor of activated T-cells, cytoplasmic 1 (NF-ATc/NFATc), is encoded by the NFATC1 gene and belongs to the NFAT family of transcription factors.
- It plays a critical role in the inducible expression of cytokine genes in T-cells, particularly driving IL-2 and IL-4 transcription upon immune activation. Related references: PMID:10358178
- NFAT2 regulates T-cell activation, proliferation, differentiation, and programmed cell death, and also controls gene expression in embryonic cardiac cells.
- The protein is essential for osteoclastogenesis, regulating key genes involved in osteoclast differentiation and bone resorption.
- Subcellular localization is dynamic: NFAT2 resides in the cytoplasm in resting cells and translocates to the nucleus upon activation/calcineurin-dependent dephosphorylation.
- Tissue expression is prominent in thymus, peripheral T-cells, and spleen; distinct isoforms (A, B, C) show preferential expression in effector vs. naive T-cells, with additional isoform IB detected in muscle, cerebellum, placenta, and brain.
- Post-translational modifications, particularly phosphorylation, control its nuclear-cytoplasmic shuttling and transcriptional activity (Keywords: Phosphoprotein, Activator, DNA-binding).
Experimental Guidance and Technical Tips
- The immunogen spans the N-terminal region (aa 10-305), present in multiple NFAT2 isoforms; this design may allow detection of various splice variants.
- For WB, consider the predicted molecular weight of ~101 kDa and validate in a relevant sample system (e.g., Jurkat T-cell lysate).
- Cross-reactivity with mouse enables use in murine models of T-cell signaling or osteoclast biology; always include appropriate controls and titrate the antibody for optimal performance.
- For ELISA, pair with a suitable detection antibody and confirm specificity using recombinant protein standards.
