Biological Background: NFAT2 Function and Localization
- NFAT2 plays a key role in the inducible expression of cytokine genes in T-cells, especially IL-2 and IL-4, and controls gene expression in embryonic cardiac cells; it regulates T-cell activation, proliferation, differentiation, and apoptosis. Related references: PMID:10358178
- The protein is essential for osteoclastogenesis and regulates many genes important for osteoclast differentiation and function.
- Subcellular localization involves shuttling between the cytoplasm and nucleus in response to calcium signaling.
- Multiple isoforms (A, B, C, IA, IB) are generated by alternative initiation and splicing, with tissue-specific expression patterns; isoforms A are enriched in effector T-cells (thymus and peripheral leukocytes) while isoforms B/C are in naive T-cells (spleen).
- NFAT2 is expressed in thymus, peripheral leukocytes (T-cells), spleen, muscle, cerebellum, placenta, and brain, with isoform-specific neural abundance in corpus callosum.
- The protein contains ANK repeats and a DNA-binding domain, and is regulated by phosphorylation; it is classified as a phosphoprotein and transcription activator/repressor.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the C-terminal region (aa 844-943) of human NFAT2, designed to be specific to this isoform and minimize cross-reactivity with other NFAT family members.
- For western blot, the predicted molecular weight is 101 kDa; consider using an 8–10% SDS-PAGE gel and optimizing transfer conditions for high-molecular-weight proteins.
- For ELISA, this antibody may be used for detection of NFAT2 in cell lysates or recombinant protein; validate the assay in your specific sample matrix.
- As a rabbit monoclonal antibody, it offers batch-to-batch consistency and high affinity; however, verify performance under your own experimental conditions.
